چکیده

Cytogenetics has proved to be a valuable aid in the study of higher organisms but has received only limited attention in microbiology. Witkin (1951), Ryan et al. (1954) and Ryan and Wainwright (1954) have successfully applied cytogenetic approaches to the study of bacteria. Haploid and diploid cultures of Escherichia coli strain K-12 were studied cytologically by Lederberg et al. (1951) in an effort to establish genetic ploidy on a cytological basis. The comparisons revealed consistent and unequivocal differences in the nuclear appearance of these two forms but failed to explain the ploidy of the cultures on a cytological basis. Clark and Webb (1955a) studied the large cells of Micrococcus aureus cytologically and radiobiologically, and presented evidence for the diploid and triploid nature of these forms. A cytogenetic study has been successfully applied to yeast by Lindegren (1949). The ploidy, life cycle, and sexual process have been substantiated by this approach although there is still some controversy in the field of yeast cvtology. The success with which the limited application of cytogeneties has met in microbiology indicates that certain of the basic problems of microbial genetics may be profitably approached by using the available tools of this field. Webb (1956) has completely reviewed the literature on cytogenetic approaches involving microorganisms. Preliminary cytological studies on Nocardia

Studies on the nutrition of Nocardia corallina.

The nutritional requirements of Streptothrix corallinus (Nocardia corallina, Breed et al., 1948) were studied by Reader (1928) who showed that growth in a medium containing glucose, ammonium sulfate and inorganic salts was stimulated by the addition of small amounts of tryptic meat broth or yeast extracts. The active principle in these additions was thought to be related to, but not identical w...

Slime as a possible factor in cell clumping in Nocardia corallina.

Work on the nuclear cytology and life cycle of Nocardia corallina has previously been reported (Webb et al., 1954; Webb and Clark, 1957; Clark and Frady, 19.57). Extensive studies on the radiation response, physiology, and genetics of this organism have also been carried ouit in this laboratory. Throughout this work, one of the main problems involved in the experimental approach has been the el...

Hydrolysis of ibuprofen nitrile and ibuprofen amide and deracemisation of ibuprofen using Nocardia corallina B-276.

A novel application of whole cells of Nocardia corallina B-276 for the deracemisation of ibuprofen is reported. This microorganism successfully hydrolysed ibuprofen nitrile to ibuprofen amide, and ibuprofen amide to ibuprofen, using a suspension of cells in a potassium phosphate buffer solution (0.1 M, pH = 7.0). These results can be explained by the presence of NHase and amidase enzymes, but t...
